The tumor cells from a primary tumor invade local tissue and gain access to the venous circulation (intravasation). Circulating tumor cells, singly or in clumps, are transported to target organs where they lodge in the capillary bed. Thus arrested, these tumor cells penetrate the endothelial cell lining and the underlining basement membrane to exit the circulation (extravasation). They then grow as a metastatic colony, a development that requires new blood vessels (neovascularization). To complete this multistep process, the tumor cells must overcome the host's defenses.
